Characterization of Three cDNAs Obtained by Spliced Leader-PCR Screening of a Taenia solium cDNA Library

چکیده
Cysticerci or metacestodes of Taenia solium cause cysticercosis, being the neurocysticercosis (NCC) main pathology. The characterization genes is essential for knowledge parasite biology, understanding parasite-host relationship, and identification possible targets diagnosis, treatment, protection. objective this work was molecular bioinformatic three news cDNAs (TsTF10, TsAAP8, TsrGAP8), obtained by spliced leader-PCR screening a cDNA library. sequences were compared with in nucleic acid protein databases (GenBank, EMBL) analyzed programs (CDD-Search National Center Biotechnology Information, Interpro European Bioinformatics Institute, Motif scan Expert Protein Analysis System Proteomics Server from Swiss Institute Bioinformatics). Considering high identities similar molecules related helminths (Taenia asiatica, Echinococcus granulosus, multilocularis, Hymenolepis diminuta) functional domains found, TsTF10, TsrGAP8 could act as nuclear transcription factor gamma, putative vacuolar ATPase membrane sector associated Rho GTPase activating protein, respectively. Although few B epitopes be predicted sequences, it relevant to evaluate them candidates diagnosis protection cysticercosis. analysis these prediction their usefulness antigens, vaccines, therapeutic contribute designing planning future studies.

Systematic screening of an arrayed cDNA library by PCR.
We have developed a PCR-based method for rapid and effective screening of arrayed cDNA libraries. This strategy directly addresses the limitations of conventional hybridization-based schemes and provides a more rapid, cost-effective, and sensitive method compatible with large-scale and routine cDNA clone recovery. To prepare arrayed libraries, 1-2 x 10(6) cDNA clones were propagated as individu...
متن کاملNested PCR for specific diagnosis of Taenia solium taeniasis.
Taeniasis due to Taenia solium is a disease with important public health consequences, since the larval stage is not exclusive to the animal intermediate, the pig, but also infects humans, causing neurocysticercosis. Early diagnosis and treatment of T. solium tapeworm carriers is important to prevent human cysticercosis. Current diagnosis based on microscopic observation of eggs lacks both sens...

Neurocysticercosis (NCC), caused by the pork tapeworm Taenia solium, is reported to be a common condition in Nepal. So far imaging diagnosis was mainstay of the diagnosis. In this paper, we report three patients presenting with neurological symptoms due to intraventricular NCC. We have diagnosed the causative agent as T. solium on molecular basis. Further research is warranted to assess the ac...
متن کاملControl of Taenia solium.
Control or eradication of Taenia solium cysticercosis has been achieved to date only in Europe and North America. Significant improvements in sanitary conditions and developing functional slaughterhouse control systems were primarily responsible for control in these regions. Conversely, in endemic areas of developing countries control is limited by economic and sanitary conditions: the life cyc...
